However, the mechanism of action of GtrS-GltR has not been definitively established. Here, we show that gltB, which encodes a periplasmic glucose binding protein, is essential for the glucose-induced activation of GtrS-GltR in P. aeruginosa. We determined that GltB is capable of binding to membrane regulatory proteins including GtrS, the sensor kinase of the GtrS-GltR TCS. We observed that alanine substitution of glucose-binding residues abolishes the ability of GltB to promote the activation of GtrS-GltR. Importantly, like the gtrS deletion mutant, gltB deletion mutant showed attenuated virulence in both Drosophila melanogaster and mouse models of infection. In addition, using CHIP-seq experiments, we showed that the promoter of gltB is the major in vivo target of GltR. Collectively, these data suggest that periplasmic binding protein GltB and GtrS-GltR TCS form a complex regulatory circuit that regulates the virulence of P. aeruginosa in response to glucose.Prophages are bacteriophages in the lysogenic state, where the viral genome is inserted within the bacterial chromosome. They contribute to strain genetic variability and can influence bacterial phenotypes. Prophages are highly abundant among the strains of the opportunistic pathogen Pseudomonas aeruginosa and were shown to confer specific traits that can promote strain pathogenicity. The main difficulty of studying those regions is the lack of a simple prophage-curing method for P. aeruginosa strains. In this study, we developed a novel, targeted-curing approach for prophages in P. aeruginosa. In the first step, we tagged the prophage for curing with an ampicillin resistance cassette (ampR) and further used this strain for the sacB counter-selection marker's temporal insertion into the prophage region. The sucrose counter-selection resulted in different variants when the prophage-cured mutant is the sole variant that lost the ampR cassette. Next, we validated the targeted-curing with local PCR amplification and Whole Genome Sequencing. The application of the strategy resulted in high efficiency both for curing the Pf4 prophage of the laboratory wild-type (WT) strain PAO1 and for PR2 prophage from the clinical, hard to genetically manipulate, 39016 strain. Future work focusing on target identification, toxicity management and reducing manufacturing time will broaden the clinical applicability and bring this exciting therapy to more patients, with longer-term remissions without additional Allo-SCT.The Barkam-Jiuzhaigou-Wuqi gravity profile extends across the Jiuzhaigou Ms7.0 earthquake (in 2017) zone and passes through several historical big earthquakes' zones. We have obtained Bouguer gravity anomalies along the profile composed of 365 gravity observation stations with Global Positioning System (GPS) coordinates, analyzed the observed data and inverted subsurface density structure. The results show that the Moho depth has a big lateral variation from southwest to northeast, which shallows from 57 km to 43 km with maximum variation up to 14 km within 800 km. The most acute depth change of the Moho is in the boundary region between the Bayan Har block and West Qinling-Qilian block. According to our analysis, it is related to the eastward movement of the Bayan Har block. There are three main pieces of evidence that support it (1) Density is higher in the east of the Bayan Har block and smaller in the west, which is the same as seismic activity; (2) Two thin low-density layers exist in the upper and middle crust of the Bayan Har block, which may promote inter-layer slip and the Jiuzhaigou Ms7.0 earthquake occurred in the boundary area of the two low-density layers, where the crustal density and Moho surface fluctuate sharply; (3) the GPS velocity field in the southwestern part gravity profile is significantly larger than that of the northeastern part, which is consistent with the density structure. Our studies also suggest that the large undulation of the Moho prevents the movement of the Bayan Har block, and strain is prone to accumulate here. The dynamic background analysis of the crust in this area indicates that the Moho surface uplifts in the West Qinling-Qilian block, which decelerates the eastern migration of material on the Qinghai-Tibet Plateau, and leads to the weak tectonic activity of the north part of the Bayan Har block.Jellyfish stings are a common issue globally, particularly in coastal areas in the summer. Victims can suffer pain, itching, swelling, shock, and even death. Usually, hot water, vinegar, or alumen is used to treat the normal symptoms of a jellyfish sting. However, a specific antivenom may be an effective treatment to deal with severe jellyfish stings. Cyanea nozakii often reach a diameter of 60 cm and are responsible for hundreds of thousands of stings per year in coastal Chinese waters. However, there has been no specific C. nozakii antivenom until now, and so the development of this antivenom is very important. Herein, we collected C. nozakii antisera from tentacle extract venom immunized rabbits and purified the immunoglobulin (IgG) fraction antivenom (AntiCnTXs). Subsequently, two complete procedures to produce a refined F(ab')2 type of antivenom (F(ab')2-AntiCnTXs) and Fab type of antivenom (Fab-AntiCnTXs) by multiple optimizations and purification were established. The neutralization efficacy of these three types of antivenoms was compared and analyzed in vitro and in vivo, and the results showed that all types of antibodies displayed some neutralization effect on the lethality of C.
